| I. Background | |
| cAMP 反应元件结合蛋白 (CREB) 位于细胞核内，是一种转录因子，在 Ser133 位点被不同的受体活化蛋白激酶（如蛋白激酶 A (PKA)、钙调蛋白依赖性蛋白激酶 (CaMK)、丝裂原活化蛋白激酶 (MAPK) 和其他激酶磷酸化后，与靶基因启动子的 cAMP 反应元件 (CRE)结合。CRE 是 CREB 的主要结合位点，负责激活 CREB。CRE 是许多细胞外和细胞内信号通路的焦点，包括 cAMP、钙、GPCR（G 蛋白偶联受体）和神经营养因子。cAMP/PKA 信号通路对许多生命过程和生物体至关重要。在 cAMP/PKA 信号通路中，CREB 顺利获得 PKA 的磷酸化被激活，并与 CRE 结合，一般基序为 5'-TGACGTCA-3'。由于 CRE 是 cAMP/PKA信号通路的调节剂，因此可以研究各种抑制剂的作用。 | |
